On the Tools menu, click E-mail Accounts.
Click View or change existing e-mail accounts, and then click Next.
In the Deliver new e-mail to the following location list, select where
you want to the new messages delivered.
Click Finish.

Yes, the new mail will be delivered to the location you specify the next time Outlook is opened and polls for mail.

Outlook cannot obtain mail unless it is running, no matter where your
delivery location is.
